Yeast aldehyde dehydrogenase. IV. Dissociation and reassociation of native and hybrid forms.
نویسندگان
چکیده
Yeast aldehyde dehydrogenase and each of two of its active proteolytic digestion products are readily dissociable into subunits which can, in turn, reassociate. The native protein dissociates into four subunits of equal sire. The partially digested enzymes dissociate into the same subunit plus a second, smaller subunit. The data are interpreted in terms of a tetrameric native enzyme, differing from its tetrameric degradation products, of which four are known, in the number of intact and functional subunits. Dissociation and reassociation of one of the hybrid forms resulted in the formation principally of a catalytically active aldehyde dehydrogenase, indistinguishable from the native enzyme.

REASSOCIATION AND REACTIVATION OF GLUCOSE 6-PHOSPHATE DEHYDROGENASE FROM STREPTOMYCES AUREOFACIENS AFTER DENATURATION BY 6 M UREA
Glucose 6-phosphate dehydrogenase (G6PD) from Streptomyces aureofaciens was purified and denatured in 6 M urea. Denaturation led to complete dissociation of the enzyme into its inactive monomers, 98% loss of the enzyme activity, about 30% decrease in the protein fluorescence and a 10 nm red shift in the emission maximum. Dilution of urea-denatured enzyme resulted in regaining of the enzyme acti...

1. Yeast alcohol dehydrogenase inactivated by reaction with iodoacetamide retains 85% of the original NADH-binding capacity as measured under conditions of saturating coenzyme concentration. 2. The dissociation constant of the enzyme-NADH complex is unaffected by inactivation of the enzyme with iodoacetamide, and the affinity of the enzyme for NAD(+) and pyridine-3-aldehyde-adenine dinucleotide...
متن کاملKinetics and reaction mechanism of potassium-activated aldehyde dehydrogenase from Saccharomyces cerevisiae.
Data from steady-state kinetic analysis of yeast K+-activated aldehyde dehydrogenase are consistent with a ternary complex mechanism. Evidence from alternative substrate analysis and product-inhibition studies supports an ordered sequence of substrate binding in which NAD+ is the leading substrate. A preincubation requirement for NAD+ for maximum activity is also consistent with the importance ...

The pH-dependent dissociation of porcine heart mitochondrial malate dehydrogenase (t-malate:NAD+ oxidoreductase, EC 1.1.1.37) has been more extensively characterized. "he native, dimeric form of the enzyme (Mr = 70,000) which exists at pH 7.5 has previously been shown to dissociate into its constituent subunits (Mr = 35,000) at pH 5.0 (Bleile, D. M., Schulz, R. A., Gregory, E. M., and Harrison,...

In this research, 11 yeast strains with ability to grow on petroleum sludge were isolated from effluent of a petroleum refinery. Based on growth on mineral media contaminated petroleum sludge, two isolates were selected as the super strains. Meanwhile, results based on biochemical and morphological experiments on the strains indicated that the two selected isolates belonged to Candida and Proto...
